Background
Protein phosphorylation is an important post-translational modification (PTM) that mediates intracellular signaling in a variety of cellular processes. More than 500 kinases encoded by the human genome are responsible for the regulation of phosphorylation events and thus are involved in many physiological and pathophysiological processes. In general, a single kinase has multiple substrates/phosphorylation sites. Although our understanding of kinase biology has improved tremendously, the specific substrates of each kinase have not been fully elucidated and these phosphorylation sites remain largely unknown. Therefore, comprehensive kinase substrate screening is necessary to understand kinase signaling networks and kinase-substrate interactions involved in disease pathology, thus contributing to the development of more specific kinase-targeted drugs.
Recent advances in mass spectroscopy and proteomics methods have enabled rapid profiling of the phosphoproteome, allowing accurate identification of phosphorylation sites and comprehensive analysis of dynamic phosphorylation events within cells. However, the identification of kinase-substrate pairs remains an exceptionally challenging task. In recent years, a variety of approaches have been continuously developed to characterize the relationship between kinases and their direct substrates and to elucidate signaling pathways. These advances in kinase substrate screening technology have the potential to transform our ability to identify new kinase targets.
