Background

Although most tissues in an organism share identical genomes and overlapping transcription profiles, each tissue exhibits diverse physiology that has important implications for both health and disease. The unique physiology of each tissue is specialized through tightly regulated gene and protein expression coordinated by differential post-translational modifications. These modifications can tune the activity of ubiquitous proteins to meet the needs of each tissue. Although physiological differences among tissues due to differential gene expression or protein abundance and subcellular localization have been revealed by genomic, transcriptomic, and proteomic analyses, they do not provide an adequate explanation for the tissue-specific effects of post-translational regulation.

Phosphorylation, a common post-translational modification, is a fundamental mechanism for regulating protein activity. It is reversible, fast, and often highly specific. Kinases and phosphatases are responsible for phosphorylation and dephosphorylation, respectively, and are themselves regulated by phosphorylation, thus forming a complex cellular signaling network. Many phosphorylation networks have been elucidated in immortalized cell lines; however, it is not sufficient to explain specialized tissue physiology. Therefore, combining large-scale, multi-tissue kinases/phosphatases expression analysis with phosphorylation site identification is essential to provide new insights into the crucial role of phosphorylation in cell signaling.
